Proper Micropipette Techniques for Accurate Laboratory Results

Why Is Pipetting Accuracy Important?

In clinical and research laboratories, accuracy in measuring liquid volumes is the foundation of every reliable test result. A pipetting error of just 1-2 microliters can cause:

  • Inaccurate test results, leading to incorrect diagnoses
  • Repeat testing that wastes reagents and time
  • Inappropriate patient care, potentially endangering health
  • Method validation failures and QC non-conformance

Therefore, choosing quality micropipettes and using proper pipetting techniques are two things that cannot be overlooked.

Understanding Micropipettes

A micropipette is a precision instrument used to aspirate and transfer liquids in micro volumes (1-1000 µL). It operates on the air displacement principle and uses disposable tips for each sample to prevent cross-contamination.

Proper Pipetting Technique (SOP)

Forward Pipetting Steps:

  1. Attach the tip properly — Press the pipette into the tip with a vertical motion and slight twist. Ensure the tip fits snugly without air gaps
  2. Pre-wet the tip — Aspirate and dispense the target liquid 2-3 times before actual measurement. This saturates the inner wall of the tip and improves accuracy
  3. Set the position — Hold the pipette vertically (90° angle) when aspirating
  4. Immerse correctly — Insert the tip 2-3 mm into the liquid (not too deep)
  5. Aspirate slowly — Press the plunger to the first stop, immerse the tip, then release the plunger slowly and consistently
  6. Dispensing — Touch the tip to the wall of the receiving vessel at a 10-45° angle, press the plunger to the first stop, then to the second stop (blow-out)
  7. Eject the tip — Use the ejector to release the used tip into a waste container
